The BY4742 strain is derived from the original strain of Saccharomyces cerevisiae - S288C. It is a commonly used laboratory strain that can be transformed with the pYES2 plasmid into BY4742 cells directly using PEG/LiAc. The selection marker for the pYES2 plasmid is URA, which can be screened using SD-URA plates. The BY4742 competent cells produced by Pointek Biotech are processed using special techniques and can be stored at -80°C for 12 months, with a transformation efficiency for the pYES2 plasmid exceeding 10⁴ cfu/μg DNA.
