M15

M15 is an engineering strain of Escherichia coli widely used in the expression of recombinant protein. Its core feature is that it carries pREP4 helper plasmid, which can constitutively express lac inhibitor, thus strictly inhibiting the background transcription of T5 promoter, which makes it particularly suitable for expressing proteins that are potentially toxic to host cells. M15 is the classic host of pQE series prokaryotic expression vectors, which have T5 promoter and N-terminal histidine tag. The strain itself is lacZΔM15 deletion type, which supports blue-white spot screening and has streptomycin resistance. PREP4 plasmid endows it with kanamycin resistance, so corresponding antibiotics should be added in culture to maintain plasmid stability. This strain usually completes plasmid transformation by heat shock method.
