LBA4404 is an Agrobacterium tumefaciens strain widely used in plant transgenic research. Its chromosome background is Ach5 type, and its nuclear gene contains rifampicin resistance gene (rifR). The auxiliary Ti plasmid carried by this strain is pAL4404, which is an Octopine plasmid. Its own T-DNA transfer function has been destroyed, but the intact vir gene segment has been retained, which can help the transformed binary vector to efficiently transfer T-DNA and integrate it into the plant genome. PAL4404 plasmid also confers streptomycin resistance (strepR) on LBA4404 strain. Therefore, rifampicin and streptomycin usually need to be added to the culture medium when the strain is used for culture or transformation screening. LBA4404 is especially suitable for genetic transformation of tobacco, tomato and other plants, and it is one of the classic tool strains in the field of plant molecular biology and biotechnology.
