EBY100 Chemically Competent Cell
EBY100 Chemically Competent Cell
The EBY100 strain is a yeast display strain developed by Invitrogen, MATa type, designed to be used with the pYD1 plasmid, allowing for direct transformation of plasmids for protein display experiments; the transformation markers are: leu2, trp1. The EBY100-pYD1 yeast display system presents target proteins on the cell surface through the a-agglutinin receptor of Saccharomyces cerevisiae, facilitating subsequent screening. The basic principle is that the a-agglutinin receptor is composed of two subunits, AGA1 and AGA2. The AGA1 protein (725 amino acids) is synthesized by EBY100 cells and secreted into the extracellular space, where it covalently binds to the β-glucan of the yeast cell wall in the extracellular matrix. The AGA2 protein (69 amino acids) can bind to the AGA1 protein via two disulfide bonds. The target protein is constructed on the pYD1 plasmid, fused with the AGA2 protein, with AGA2 at the N-terminus and the target protein at the C-terminus. When the EBY100 yeast containing the pYD1 plasmid is induced to express in a medium containing galactose and lacking glucose, it can express the AGA2-target protein fusion protein. This fusion protein binds to the AGA1 protein on the surface of EBY100 yeast cells via AGA2, thus displaying the target protein on the yeast surface. The EBY100 competent cells are prepared through a special process and can be stored at -80°C for 12 months, while the pYD1 plasmid shows a transformation efficiency greater than 104 cfu/μg DNA.
