AE(DE3), namely, ArcticExpress (DE3), originated from E. coli B, is a strain deficient in Lon and OmpT protease, which can promote the stability of the expressed protein. The chromosome DNA of AE (DE3) strain integrates the DE3 region of λ phage, which enables AE (DE3) strain to simultaneously express T7 RNA polymerase and Escherichia coli RNA polymerase, and is widely used for protein expression of pET series, pGEX, pMAL and other plasmids. AE (DE3) strain is resistant to tetracycline and gentamicin, and endA1 mutation is beneficial to the stability of plasmid DNA. The existence of [cpn10cpn60 GentR] enables AE (DE3) to express chaperones Cpn10 and Cpn60 (from the psychrophilic bacterium —Oleispira antarctica) adapted to low temperature. Cpn10 and Cpn60 chaperones show high activity at 4-12℃. When expressed in AE(DE3) cells, they can reduce the formation of inclusion bodies of recombinant proteins, increase the expression amount and biological activity of soluble recombinant proteins, and have better fusion-promoting properties than traditional prokaryotic expression chaperones GroEL and GroES.
