Pink1
Pink1
Pink1 yeast strain (also known as PichiaPink Strain 1) is an engineering strain of Pichia pastoris, which was developed by Invitrogen Company and mainly used for eukaryotic expression of recombinant protein. Its core feature is the deletion of ADE2 gene, which leads to two key results: first, the strain becomes adenine auxotroph (Ade2-) and cannot grow on the medium without adenine; The second is the accumulation of purine precursors in cells, which makes colonies appear pink on conventional YPD medium. When successfully transferred into a matching plasmid (such as pPinkα-HC) containing ADE2 screening marker, the color of transformant will change from pink to white, and it can grow on the medium lacking adenine. This intuitive color change provides a convenient sign for the screening of positive transformants. This strain is usually transformed by electroporation, which is a common tool for protein expression research.
