Pink2
Pink2
Pink2 yeast (officially named PichiaPink Strain 2) is an engineering strain of Pichia, which was developed by Invitrogen Company and mainly used for eukaryotic expression of recombinant protein. Its core feature is the deletion of ADE2 gene, which leads to the strain becoming adenine auxotrophic and forming pink colonies on conventional YPD medium. When successfully transferred into the plasmid containing ADE2 marker gene (such as pPinkα-HC), the transformant can grow on the medium without adenine, and the colony color turns white, which provides convenience for the screening of positive clones. In addition, the strain also knocked out PEP4 gene, and its encoded proteolytic enzyme A is a key regulator of various protease activities in vacuoles. This defect can effectively reduce the degradation of foreign expressed protein by host protease, which is helpful to improve the yield and stability of target protein.
