EGY48(p8op-lacz) Chemically Competent Cell

The EGY48(p8op-lacz) strain is a yeast strain for use in the LexA system two-hybrid experiment, MATα type, and can directly transform plasmids to verify protein interactions or screen libraries; the transformation markers are: his3, trp1, ura3, and the reporter gene is: LEU2; the reporter gene UAS (upstream activating sequence) is derived from LexA op(x6), and LEU2 expression can only be activated when Bait and Prey interact. The EGY48-LexA system includes three corresponding plasmids: pLexA, pB42AD, p8op-LacZ. The selection marker for plasmid pLexA is HIS3, used for expressing the fusion protein of DNA-BD and the target protein (Bait); the selection marker for plasmid pB42AD is TRP1, used for expressing the fusion protein of AD and the target protein (Prey); the selection marker for the reporter plasmid p8op-LacZ is URA3, with the reporter gene LacZ, and the reporter gene UAS also comes from LexA op(x6), which can only activate LacZ expression when Bait and Prey interact. EGY48(p8op-lacz) competent cells are prepared using a special process, and the transformation efficiency for the pGBKT7 plasmid is >10⁴ cfu/μg DNA.
