SMD1168H
SMD1168H
SMD1168H is a Pichia pastoris strain, belonging to eukaryotic cells. Generally, antibiotics such asanamycin and ampicillin, which are effective against prokaryotes, are ineffective against yeast. Therefore, in order to prevent contamination of yeast culture strains by proaryotes such as E. coli, some antibiotics such as ampicillin and kanamycin are often added to the culture medium to inhibit the contamination and growth of strains. The suitable growth temperature for Pichia pastoris SMD1168H is 28-30℃, and 30℃ is generally used culture, and temperatures above 32℃ may lead to cell death. A mutation in the pep4 gene of the Pichia pastoris SMD1168H genome results in the loss of activity of the protease A, which can protect the expressed product from degradation and promote the increase in expression, and the resulting transformant is the Mut (methanol utilization plus) genotype. Pichia pastoris SMD1168H is recommended for the expression of recombinant plasmids containing Zein resistance, and the working concentration of Zeocin antibiotic is recommended to be 100μg/ml when screening for SMD1168H recombin transformant strains. Pichia pastoris SMD1168H genotype: pep4.
