R5421
R5421
R5421 is a competent cell of Saccharomyces cerevisiae for the study of potassium ion transport. The genotype is mat α ura3-52 leu2 trk1 δ his3 δ 200 his4-15 trk2 δ 1:: pck64. The strain is potassium deficient, its growth is highly dependent on the concentration of potassium ion in the culture medium, and it stops growing when it is lower than 0.5mM KCl, so it is specially used for functional identification of potassium ion channels, transporters or sodium-potassium pumps. The transformation screening markers were ura3 and leu2, and the transformation efficiency of pGADT7 plasmid was more than 10 cfu/μ g DNA. The strain was sensitive to high temperature, and the optimum culture temperature was 27-30℃.
