BTH101 is an Escherichia coli engineering strain widely used in bacterial two-hybrid system. Its key feature lies in the deletion of cyaA gene encoding adenylate cyclase, which leads to extremely low intracellular cAMP level and can not activate genes related to glucose metabolism. When the proteins to be detected interact with each other and reconstruct the signal pathway of cAMP, red colonies can be formed on MacKay plate or blue color reaction can be produced on X-gal plate. The strain is suitable for verifying protein interaction in vivo.
