AXT3K Chemically Competent Cell
AXT3K Chemically Competent Cell
![]() The AXT3K strain originates from the YSH yeast strain, which is of the MATa type. The transformation markers are ura3 and trp1, making it suitable for selecting plasmids marked URA3 and TRP, such as pYES2/NT, pYC2/NT, pDR196, p416GDP, pESC-URA, and pESC-TRP. The wild-type Saccharomyces cerevisiae has the ability to absorb and transport sucrose. The AXT3K strain is a sucrose non-fermenting (invertase-deficient) strain that cannot grow on media with sucrose as the sole carbon source, making it an efficient experimental platform for the discovery and study of natural or heterologous (from other species) sucrose transport proteins. New sucrose transport proteins can be identified through functional complementation assays. The AXT3K strain grows normally in media containing 2% glucose, and it is recommended to culture AXT3K yeast cells using YPD/YPDA medium; for selecting AXT3K yeast cells containing plasmids, SD medium is recommended. The competent AXT3K cells are prepared using a special process and can be stored at -80°C for 12 months. The transformation efficiency of the pYES2 plasmid (5857 bp, AmpR) is measured to be >104 cfu/μg DNA.
