THY.AP5 Chemically Competent Cell
THY.AP5 Chemically Competent Cell
The Split Ubiquitin System (mbSUS) is a detection technology specifically designed to screen for interactions between transmembrane proteins. It utilizes a split ubiquitin system to directly detect interactions between membrane proteins in their native state through a yeast two-hybrid system. This system employs the THY.AP5 yeast strain, which allows for the direct transformation of plasmids for protein interaction validation or library screening experiments. The system also utilizes THY.AP4 and THY.AP5 yeast strains, where the genotype of the THY.AP5 strain is: MATα; URA3; ade2-, his3-, leu2-, trp1-. When used in combination with THY.AP4, selective growth screening is first conducted through auxotrophic reporter genes (HIS3, ADE2), followed by β-galactosidase analysis using the LacZ reporter gene for quantitative or semi-quantitative screening. Three independent reporter genes regulated by different promoters help to reduce the likelihood of false positives. The system can use plasmids such as: pNXgate32-3HA, pXNgate21-3HA, pMetYCgate. THY.AP5 competent cells are specially prepared and can be stored at -80°C for 12 months, with the pGADT7 plasmid achieving a transformation efficiency of >104 cfu/μg DNA.
