△smf Chemically Competent Cell

The △smf strain is derived from the BY4741 yeast strain, which has a deleted endogenous gene △smf that has a high affinity for manganese ions, significantly reducing its ability to absorb and transport manganese ions. △smf is often used in assays to identify manganese ion transport proteins. This strain can grow normally on basic medium (catalog number: PT1072) but grows poorly on media containing high concentrations of the divalent cation chelator EGTA. pDR196 is a high-efficiency expression vector for Saccharomyces cerevisiae proteins, with a selection marker of Ura3. The gene X encoding the manganese ion transport protein is constructed into the pDR196 vector (pDR196-X), and transformed into △smf competent cells, where the X protein will mediate the uptake of manganese ions by the yeast cells, enabling △smf yeast to grow normally in a medium lacking manganese ions.
