△zrc1 cot1 Chemically Competent Cell
△zrc1 cot1 is mainly used to detect protein zinc transport function by yeast complementation experiment. It is an engineered strain of bre's yeast in which both ZRC1 and COT1 genes are deleted simultaneously. Both genes encode zinc ion transport proteins (mainly localized in the vacuolar membrane) and play a key role in maintaining the zinc ion homeostasis in cells. Their double deletion is predicted to cause severe damage to the ability of yeast cells to store zinc in vacoles, which may show a high sensitivity to zinc ion toxicity or imbalance of zinc distribution in cells. This kind of strains are usually used to study the homeostasis of metal ions the mechanism of metal toxicity tolerance and related human disease models. △zrc1 cot1 competent cells were made by special process, which can be stored at -80 for 12 months, and the transformation efficiency was tested by pYES2 plasmid (5857bp, AmpR) >104 cfu/μg DNA.
