SMD1163 Chemically Competent Cell

SMD1163 is a strain of Pichia pastoris, which belongs to eukaryotic cells. Common antibiotics targeting prokaryotes, such as kanamycin and ampicillin, are ineffective against yeast; therefore, to prevent contamination of yeast cultures by prokaryotic organisms like Escherichia coli, antibiotics such as ampicillin and kanamycin are often added to the culture medium to inhibit bacterial contamination and growth. The optimal growth temperature for SMD1163 Pichia pastoris is 28-30°C, typically cultured at 30°C, and temperatures exceeding 32°C may lead to cell death. SMD1163 Pichia pastoris is a histidine auxotrophic strain and cannot grow in minimal media lacking histidine. The compatible vectors for SMD1163 are yeast expression vectors containing the His4 gene, such as pPIC9K and pPIC3.5K. The Pep4 and prb1 genes in the SMD1163 genome have mutations that lead to the loss of activity of proteinase A and proteinase B, which protects the expression products from degradation and promotes an increase in expression levels. The SMD1163 strain is of the Mut type, capable of producing alcohol oxidase and normally utilizes methanol.
