EPI400

EPI400 is an engineering strain of Escherichia coli for cloning unstable DNA or toxic genes. Its key feature is that pcnB gene is regulated by inducible promoter, and the plasmid copy number is low when WDEPI inducer is not added, which is beneficial to stable cloning. After induction, the high copy number can be recovered to extract a large number of plasmids. This strain is suitable for cloning DNA rich in methylated cytosine or adenine, and supports blue and white spot screening. The transformation efficiency of competent cells is high, for example, it can reach 5 × 10 cfu/μ g DNA by pUC19 detection.
