OrigamiB(DE3)
OrigamiB(DE3)
OrigamiB(DE3) is an engineering strain of Escherichia coli used to express active protein rich in disulfide bonds. The strain originated from BL21, and its key feature lies in the simultaneous mutation of thioredoxin reductase (trxB) and glutathione reductase (gor) genes, which makes its cytoplasmic environment more conducive to the correct formation of disulfide bonds, thus significantly improving the soluble expression of complex active proteins. The chromosome of the strain is integrated with λDE3 lysosome, which is suitable for IPTG-induced expression of T7 promoter vectors such as pET series. TrxB and gor mutations give kanamycin and tetracycline resistance, respectively, so it can not be used in plasmids with these two resistance markers. The strain integrates the defect characteristics of lon and ompT protease of BL21, which is helpful to enhance the stability of the expressed protein.
