TSsetta(DE3)
TSsetta(DE3)
TSsetta(DE3) is an Escherichia coli engineering strain commonly used for protein expression. Its core feature is that it can supplement the tRNA corresponding to a variety of rare codons that Escherichia coli lacks, thus effectively improving the expression level of foreign genes such as eukaryotic sources. The strain has integrated the DE3 region of λ phage on its chromosome, and contains T7 RNA polymerase gene controlled by lacUV5 promoter, which can be used for induced expression of T7 promoter vectors such as pET series, pGEX and pMAL. The pRARE2 plasmid (with chloramphenicol resistance) can provide tRNA for six rare codons: AUA, AGG, AGA, CUA, CCC and GGA. TSsetta(DE3) competent cells are usually transformed by heat shock, which is suitable for efficient expression of recombinant protein.
