HB101 is a hybrid derivative of Escherichia coli K-12 and B strain, and it is a classic tool for early molecular cloning. Its key characteristics include: recA13 mutation makes it lack homologous recombination activity, which is conducive to the stable cloning of repeated sequences or large fragments of DNA；; Restriction system deletion (hsdS20) can significantly improve the transformation efficiency of foreign DNA; At the same time, it is thymidine kinase deficiency (thyA). This strain is suitable for plasmid amplification and library construction, but it should be noted that it does not contain F' factor and cannot be used for filamentous phage infection experiments.
