S17-1 λpir
S17-1 λpir
S17-1 λpir is an Escherichia coli engineering strain commonly used in conjugation transfer experiments. Its key feature is that some functional fragments of RP4-2 conjugation transfer plasmid are integrated in chromosome, which can mediate DNA transfer between different strains independently and complete the conjugation process without auxiliary strains. The strain is recombinase deficient (recA1), which can reduce homologous recombination of DNA and ensure the stability of foreign fragments. At the same time, the deletion of endonuclease (endA1) is helpful to improve the quality and yield of plasmid extraction. In addition, the strain carries λpir element, which allows plasmids (such as some suicide vectors) that rely on π protein replication to exist stably in the bacteria. The transformation efficiency of competent cells detected by pUC19 can reach 108 cfu/μg DNA, which is suitable for experimental operations such as gene cloning and conjugation transfer.
