OverExpress C43(DE3) is an engineering strain of Escherichia coli derived from BL21(DE3), which is designed for efficient expression of toxic proteins and hydrophobic membrane proteins. At least two unknown mutations were obtained by phenotypic screening, which enabled the strain to tolerate cell death caused by the expression of toxic proteins, thus significantly improving the soluble yield of complex recombinant proteins. Its genotype is F- ompT hsdSB (rB- mB-) gal dcm (DE3), and the chromosome integrates the DE3 region of λ phage, which is suitable for IPTG-induced expression of T7 promoter vectors such as pET and pGEX. The transformation efficiency of chemocompetent cells of the strain is high, for example, it can reach the order of 108 cfu/μg DNA detected by pUC19, which is widely used in protein expression research.
