Origami2(DE3)
Origami2(DE3)
The Origami2(DE3) strain is a derivative of K-12 containing mutations in the thioredoxin reductasetrxB) and glutathione reductase (gor) genes, which are two key enzymes in the reducing pathway. The mutation is beneficial for the correct folding of proteins disulfide bonds, enhancing protein solubility. Meanwhile, this strain does not have kanamycin resistance, which can be used for protein expression with plasmids that kanamycin resistance. The Origami2(DE3) strain chromosomally integrates the λ phage DE3 region (which contains the T7 phage RNA polymer), and this region is integrated into the E. coli chromosome, allowing for the simultaneous expression of T7 RNA polymerase and E. coli RNA polymerase, which be used for protein expression in pET series, pGEX, pMAL and other plasmids. The Origami2(DE3) strain has streptomy, tetracycline resistance, and is a leucine auxotrophic strain.
