This kit is specially designed for tender true leaves of six-leaf-stage pepper seedlings. It contains all necessary components, including enzymatic digestion solution, washing solution, resuspension solution, and transformation reagents. Based on a 5 mL enzymatic digestion system, it can be used for 40 preparations and comes with consumables such as a 70 μm cell strainer. Store at -20°C, with a shelf life of one year. Utilizing an optimized enzymatic digestion formula, combined with peeling or finely cutting treatment, the yield of protoplasts can reach up to 14.70×10⁶/g FW, with viability over 88%. Relying on an improved transformation system, the transformation efficiency is increased to over 75%, far exceeding traditional methods. The prepared protoplasts can be directly used for research such as protein localization and interaction verification. The operation process is clear and requires no complex equipment, providing an efficient tool for molecular biology research in pepper.
