Tea Leaf Protoplast Preparation and Transformation Kit Plus

This kit is specially designed for the leaves of tea seedlings at the 3–4 true leaf stage. It contains a complete set of components including enzyme digestion solution, washing solution, MMg solution, and PEG transformation solution, along with consumables such as a 70μm cell strainer. Store at -20°C, with a shelf life of one year. The optimized formulation contains 1.5% cellulase, 2.5% pectolyase, and 0.6 mol/L mannitol. After enzyme digestion in darkness at 25°C for 16–18 hours, the yield and viability of protoplasts are excellent. Based on the improved transformation system, combined with an iodixanol purification step, the transformation efficiency is ensured. The prepared protoplasts can be directly used for studies such as protein localization and gene expression regulation. The operation process is clear and does not require complex equipment, providing an efficient tool for molecular biology research in tea plants.
