Ruby Visualization Protein Interaction Verification Kit

The Gal4-VP16-UAS system: The Gal4-VP16 fusion protein (Gal4: binding domain; VP16: activation domain) can bind to the UAS sequence element, thereby initiating the expression of the downstream reporter gene. When the two domains are expressed separately, their spatial proximity enables the formation of a complete protein, which binds to the UAS and activates the downstream reporter gene expression.
Ruby: The Ruby reporting system demonstrates the activity of three enzyme proteins (CYP76AD1, DODA, and GT), which catalyze the degradation of tyrosine substrate into visible rose-red betaxanthin. As tyrosine is a natural plant substrate, no additional substrate addition is required for in situ colorimetric detection.

uvGFP: Under 365 nm excitation light, uvGFP emits visible green fluorescence that is dozens of times stronger than eGFP, requiring no filter for observation.
Interaction validation: The ruby reporter gene is flanked by a 5xUAS sequence. Protein A is fused and expressed with Gal4, while protein B is fused and expressed with VP16. If A and B interact to form the complete Gal4-VP16 protein, this complex binds to the 5xUAS sequence, initiating downstream ruby gene expression and producing visible rose-red pigment. Additionally, the reporter vector contains an uvGFP expression cassette. The uvGFP gene is preceded by a strong constitutive promoter, serving as an internal reference for verifying successful transient transformation.
