The TPS2 gene encodes “trehalose-6-phosphate phosphatase”. Like the TPS1 gene, it also a key enzyme in the yeast cell's trehalose synthesis process and is also the main rate-limiting step in trehalose biosynthesis. In the twostep synthesis pathway of trehalose, the enzyme encoded by the TPS2 gene is responsible for the “second reaction”. Specifically, it will “dephosphorylate trehalose-6-phosphate” after the “trehalose-6-phosphate synthetase (encoded by the TPS1 gene)” cataly the generation of trehalose-6-phosphate, and ultimately generate trehalose. The Δtps2 yeast mutant cannot grow on glucose or fructose and rapidly fermentable carbon sources, but can grow normally on glycerol or ethanol and other non-fermentable carbon sources.
