Ruby: The Ruby reporter system detects three enzyme proteins (CYP76AD1, DODA, and GT), which catalyze the degradation of the substrate tyrosine to produce visible rose-red betaxanthin. For the Dual-LUC reporter system, the substrate must be added and its expression level detected using a chemiluminescence microplate reader. In contrast, tyrosine serves as a natural substrate in plants, enabling direct in situ colorimetric detection without artificial substrate addition.
uvGFP: Under a 365 nm excitation light source, uvGFP emits visible green fluorescence that is dozens of times stronger than eGFP, requiring no filters for observation.
The DNA sequence to be verified is constructed upstream of either the ruby reporter gene (with uvGFP serving as the internal reference, activated by a strong promoter) or the uvGFP reporter gene (with ruby serving as the internal reference, activated by a strong promoter), using the pGreenII-SK vector to express the transcription factor. When the transcription factor binds to the DNA, it initiates the expression of the downstream reporter gene. The interaction is detected by observing color (red fluorescence from ruby expression) or fluorescence (green fluorescence from uvGFP expression). This system is primarily used for qualitative analysis.
