L40
L40
L40 is a Saccharomyces cerevisiae reporter strain used in yeast two-hybrid system. Its genotype is MATa, and it carries trp1, leu2, his3 and other auxotrophic markers. Two reporter genes, HIS3 and lacZ, which are regulated by LexA operator, are integrated in the genome of this strain. When bait protein interacts with prey protein, the expression of reporter gene can be activated, so that the protein interaction can be verified by the growth status and β -galactosidase activity detection on histidine-deficient medium. L40 is suitable for direct co-transformation of plasmids for interaction verification, and it is a classic tool for studying protein interactions.
