YTK12
YTK12
YTK12 is a saccharase exocrine deficient Saccharomyces cerevisiae, which is widely used to analyze and identify signal peptides of secreted proteins. Sucrase transformation gene SUC2 can transform raffinose or sucrose into glucose that can be used by yeast. pSUC2 vector used in yeast signal peptide tracking system contains suc2, but it is a gene with no secretory activity and no signal peptide sequence. YTK12 yeast strain is a suc2 gene deficient strain, which can't grow normally on YPRAA medium with raffinose as a single carbon source. Only by connecting the signal peptide sequence with secretory activity to the N-terminal of suc2 gene of pSUC2 vector and transforming it into YTK12 yeast strain can sucrose invertase be secreted normally, and YTK12 yeast can transform raffinose into glucose and grow normally on YPRAA medium (Jacobs et al 1997, Oh et al 2009).At the same time, color reaction can be used to further verify the results, because the enzymatic product of SUC2 can reduce 2,3,5- triphenyltetrazolium chloride (TTC) to insoluble red 1,3,5- triphenylformic acid (TPF), which has been widely used to study the function of signal peptide.
