The genotype of THY.AP5 strain is: MATα; URA3; ade2-, his3-,leu2-, trp1-。 When used in combination with THY.AP4, the first step is selective growth screening by auxotrophic reporter gene (HIS3, ADE2), and then quantitative or semi-quantitative screening by LacZ reporter gene for β -galactose analysis. Three independent reporter genes are regulated by different promoters, which reduces the false positive rate. The plasmids used in this system are: pNXgate32-3HA, pXNgate21-3HA, pMetYCgate.
