△PMC1 VCX1 is an engineering strain of Saccharomyces cerevisiae with double gene deletion, which was constructed by simultaneously knocking out pmc1 and vcx1 genes. PMC1 encodes a P-type calcium pump located in vacuole membrane, which is responsible for actively transporting calcium ions in cytoplasm to vacuole for storage; VCX1 encodes an H+/Ca2+ exchange protein located in the vacuole membrane, and also participates in the storage of calcium ions in vacuoles. Double deletion leads to serious damage to the calcium buffering capacity of yeast cytoplasm, which is extremely sensitive to high calcium environment, and the vacuole calcium storage function is significantly reduced. This strain is an extremely sensitive model for studying calcium ion homeostasis, calcium signal transduction and vacuole calcium storage function.
