SM10λpir
SM10λpir
SM10λpir is an Escherichia coli engineering strain specially designed for indirect bacterial transfer. Its core feature is that the chromosome is integrated with the junction transfer (tra) gene of RP4 plasmid, which can autonomously start the transfer of DNA to other strains. The strain is recombinant defective (recA), which ensures the stability of foreign DNA. It carries λpir elements, allowing suicide vectors (such as pDM4) that rely on π protein replication to exist stably. SM10λpir is suitable for transferring plasmids from Escherichia coli to Gram-negative bacteria which are difficult to be directly transformed, such as Agrobacterium, and is a common tool for constructing gene knockout mutants.
