Rosetta 2(DE3)
Rosetta 2(DE3)
Rosetta 2(DE3) strain is derived from BL21, and it is a defective strain of Lon protease and extracellular protease OMPT. The deletion of these two enzymes effectively prevents the degradation of heterologous proteins in Escherichia coli. Introducing pRARE2 plasmid with chloramphenicol resistance into BL21 (DE3) cells is Rosetta 2(DE3). pRARE2 plasmid can supplement tRNA corresponding to seven rare codons (AUA, AGG, AGA, CUA, CCC, GGA and CGG) lacking in E.coli, and improve the expression level of foreign genes. The chromosome of the strain is integrated with the DE3 region of λ phage (the DE3 region contains T7 phage RNA polymerase), which can simultaneously express T7 RNA polymerase and Escherichia coli RNA polymerase, and can be used for protein expression of pET series, pGEX, pMAL and other plasmids.
