△Pep4 Yeast strain
△Pep4 Yeast strain
The PEP4 gene encodes protease A, a key aspartic protease in vacuoles. Deletion of the Pep4 gene reduces the activity of various vacuolar hydrolases, including protease B—which relies on protease A for maturation—by 90–95%. This significantly decreases the degradation of heterologously expressed proteins and enhances yield. Due to impaired proteolytic capacity, the mutant exhibits impaired autophagy (autophagosomes fail to be effectively degraded within vacuoles), although growth in conventional culture media remains essentially normal.
