Soluble starch medium
Soluble starch medium is a differential medium used to detect the activity of microbial amylase. Its basic components provide nitrogen source for peptone and beef extract powder, and the key additive is soluble starch as enzymolysis substrate. After culture, Lugo's iodine solution was added dropwise, and the transparent hydrolysis circle appeared around the amylase-positive colony because the starch was hydrolyzed, while the unhydrolyzed area was blue-black. The yield of amylase can be evaluated by measuring the ratio of hydrolysis circle to colony diameter, which is an important tool for screening enzyme-producing strains.
