ClearColi BL21（DE3）Escherichia coli strain

ClearColi BL21 (DE3) is derived from BL21 (DE3) and carries eight independent gene mutations that modify the lipopolysaccharide (LPS) in the outer cell wall of ClearColi BL21 (DE3). The oligosaccharide chains of LPS are deleted, along with two acyl chains, thereby disrupting the endotoxin signaling pathway in ClearColi BL21 (DE3). Normal LPS contains six acyl chains; LPS with six acyl chains can be recognized by the receptor TLR4, triggering the endotoxin response, whereas LPS with only four acyl chains cannot be recognized by TLR4 and thus does not initiate the endotoxin response. The endotoxin content in proteins or plasmid DNA extracted from ClearColi BL21 (DE3) cells is extremely low, making the endotoxin-free proteins or peptides widely applicable in mammalian experiments and disease treatment. ClearColi BL21 (DE3) is also a defective strain for both Lon protease and the extracellular membrane enzyme OMPT; the absence of these enzymes effectively prevents the degradation of heterologous proteins within Escherichia coli, enhancing the stability and expression levels of target proteins. The λ phage DE3 region contains T7 phage RNA polymerase, which is integrated into the chromosome of ClearColi BL21 (DE3) and enables simultaneous expression of both T7 RNA polymerase and E. coli RNA polymerase for protein expression on vectors such as pET, pGEX, and pMAL.
