10×TE/LiAc Solution

This product is a 10×TE/LiAc mixture composed of 100 mM Tris, 10 mM EDTA, and 1 M LiAc, which has undergone filtration and sterilization. It can be prepared as 1.1× TE/LiAc or PEG/LiAc solutions for the preparation and transformation of Saccharomyces cerevisiae competent cells. The Li⁺ ions in LiAc alter cell membrane permeability, promoting the formation of competence and facilitating the uptake of exogenous DNA by cells. PEG protects the cell membrane in high-concentration LiAc environments, reducing excessive membrane damage caused by LiAc while enhancing tight contact between plasmids and the cell membrane. The TE buffer provides a stable pH environment.
