pML105 Vector
pML105 Vector
The pML105 vector is based on the pML104 scaffold but replaces the Ura selection marker with Trp, belonging to the CRISPR/Cas9 gene-editing system for Saccharomyces cerevisiae. Its core structure comprises the Cas9 nuclease encoding gene (driven by a GAP promoter) and a single guide RNA (sgRNA) expression cassette (regulated by a SNR52 promoter). By inserting a guide sequence targeting specific genes into the sgRNA expression cassette, Cas9 can be directed to induce double-strand breaks at designated sites in the S. cerevisiae genome, thereby introducing genetic mutations via non-homologous end joining (NHEJ) or homologous recombination (HR). This vector is suitable for site-specific mutation, gene knockout, and base editing in functional studies of S. cerevisiae genes.
