pGK415-rtetTAK8N, L131L Vector
pGK415-rtetTAK8N, L131L Vector
The plasmid pGK415-rtetTAK8N, L131L was constructed by the team of Jun Ishii at Kobe University in Japan and published on Addgene (plasmid number #165969). It is a plasmid designed for gene-induced expression in yeast. The expressed fusion protein (rtTA) does not normally bind to DNA. Upon addition of doxycycline (Dox) to the culture medium, rtTA undergoes conformational changes, binds to the promoter containing the tetO operon sequence, and activates downstream gene transcription via its VP16 domain. Consequently, this vector is widely utilized to construct "drug-on" inducible expression systems for investigating the function or toxicity of specific genes.
