YB525 Yeast strain
YB525 Yeast strain
YB525 is a widely used mutant strain of Saccharomyces cerevisiae for fatty acid metabolism research, characterized by the knockout of two major long-chain acyl-CoA synthetase genes, FAA1 and FAA4. Consequently, it cannot activate exogenous long-chain fatty acids and fails to grow normally on media containing only fatty acids (e.g., C16:0, C18:0) as the sole carbon source, which serves as the foundation for its use as a functional screening platform. This strain is commonly employed for heterologous expression of LACS genes from various species; growth recovery can be observed to verify gene function or analyze enzyme substrate preferences. Its genotype is: MATa (can1-100 ade2-1 his3-11,15 leu2-3,112 trp1-1 ura3-1 faa1△faa4△).
