pINA1269 is a single-copy integrated expression plasmid specifically designed for Yarrowia lipolytica, and it is one of the most widely used integrated vectors in metabolic engineering and heterologous protein expression studies in this organism. This plasmid is integrated into the yeast genome as a single copy via homologous recombination, enabling stable and heritable expression of exogenous genes while avoiding the loss of free plasmids in the absence of selective pressure. It contains a constitutive protein expression cassette consisting of two key elements: the strong constitutive promoter hp4d, which serves as the core component driving high-level expression of exogenous genes in Yarrowia lipolytica, and the XPR2 terminator, derived from the transcription termination signal of the alkaline extracellular protease gene, ensuring proper termination of exogenous gene transcription.
