Bacterial Single Hybrid Interaction Verification Kit
Bacterial Single Hybrid Interaction Verification Kit
Our company has developed a bacterial single-hybrid validation kit that leverages the in vivo environment of Escherichia coli as a tool for studying DNA–protein interactions and for identifying inhibitors targeting specific transcription factor–DNA interactions. The core principle involves placing the DNA sequence under investigation (the bait) upstream of a reporter gene; when it specifically binds to a co-expressed transcription factor (the prey), it activates the expression of the His3 and aadA reporters (streptomycin resistance, which is not commonly used), thereby allowing the interaction to be determined through a bacterial survival assay. In this protocol, the optimal concentration of 3-AT for detecting the bait Escherichia coli strain and the validation of interactions were both performed using the dilution-point plate method. Compared with the spread-plating method, the point-plating method provides a more intuitive visualization of DNA–protein interactions and reduces the amount of culture medium and 3-AT required. This protocol also includes an analysis of common bacterial single-hybrid results.
